Phosphothioate Oligonucleotide

PTO

PTO (Phosphothioate Oligonucleotide) contain one sulfor atom in place of an oxygen atom in the internucleotide linkage of DNA or RNA. Phosphothioate oligonucleotide can be ordered as

  • Full PTO (whole oligo contains phosphothioate linkages)
  • “Thiocap” PTO (only a few phosphothioate linkages at the 3′ or 5′-end)
  • Chimera (mixed RNA/DNA with stretches of phosphothioate)

The more phosphothioate linkages present in the oligonucleotide, the higher the stability. Phosphothioate oligonucleotides are useful as antisense molecules inhibiting gene expression because of their resistance to nuclease degradation.
